1. 250 nm is in the UV region of light, and the spectrometer is often unable to detect this region of light and result in appearing having no transmittance. 2. Since most biological macromolecules (protein in your case) do not absorb light in the visible region (~99% transmittance) but only absorb in the UV region, so the concentration can be determined from the corresponding absorbance in the UV region of light.
